After five players withdrawing from the England camp prior to Saturday's game, the major talking point from the starting XI was the inclusion of goalkeeper James Trafford ahead of usual number one Jordan Pickford.
Playing in their first home game since finishing third at the World Cup but ultimately suffering semi-final heartbreak, Thomas Tuchel's side made the worse possible start as Marc Guehi was dispossessed and Lamine Yamal took full advantage to give Spain a second minute lead.
Spain created a number of chances and World Cup final hero Ferran Torres missed two glorious opportunities to make it 2-0 as he was denied by Trafford and then missed the target when through on goal minutes later.
The World Cup champions initially regretted their missed opportunities as the hosts suddenly burst into life on 36 minutes.
An excellent counter attack ended with Jude Bellingham providing the perfect pass for Anthony Gordon to slot home past Unai Simon.
Just over three minutes later and England had the lead when a terrific cross from Nico O’Reilly was headed home by Harry Kane via a Marc Cucurella deflection.
After an absorbing first half, the Three Lions were presented with the perfect chance to double their lead when Bellingham was fouled by Rodri in the box.
Usually so reliable from 12 yards, Kane's strike crashed the underside of the bar and he was penalised for a double hit after falling when he took the penalty kick.
An end-to-end encounter continued and this time it was O'Reilly's turn to be punished as he gave the ball away in a dangerous area and Alex Baena took full advantage to slot home and make it 2-2 on the hour mark.
After both sides missed chances to retake the lead, it was Spain substitute Mikel Oyarzabal who provided the match-winning moment as his wonderful finish ensured Spain extended their incredible unbeaten run to 39 games and made a winning start to the 2026/27 UEFA Nations League.
While there was plenty for Tuchel to like about the way his side played as an attacking force, he will be hoping his team can cut out the defensive mistakes when they play there next three games before the end of the international break.
