The NFL heads Down Under for the first time early Friday morning as San Francisco 49ers take on Los Angeles Rams at the Melbourne Cricket Ground.

A sellout stadium full of expectant fans will hope to see two genuine Super Bowl contenders produce a memorable Week 1 contest.

The Niners decamped to Australia earlier this week in a bid to acclimatise gradually, while the Rams fly in as late as possible to lessen the inevitable body-clock disruption.

Both teams reached the play-offs last season before losing to NFC West rivals the Seattle Seahawks.

Los Angeles took a big swing at improving their defence this offseason with a trade for single-season sack king Miles Garrett and the return of Super Bowl winner Aaron Donald.

The defensive tackle is not available for this game as he ramps up his comeback bid while Fred Warner, Nick Bosa and George Kittle are set to return from injury for the 49ers, with Mike Evans making his debut after arriving via free agency.

Both teams are loaded with star power and both are desperate to set the tone for what promises to be a ferocious NFC West race.

Ten touchdowns were scored the last time these two teams met in November, with the Rams prevailing 42-26 against their injury-hit division rivals.

Mac Jones filled in at quarterback for Brock Purdy and passed for over 300 yards and three TDs, but the Rams' Matt Stafford eclipsed him with four scoring strikes, all to different receivers.

San Francisco were without their two top defensive stars Bosa and Warner, plus first-round rookie defensive end Mykel Williams, so Stafford enjoyed a level of comfort in the pocket that might not be repeated.

But the Rams' receiving corps remains one of the best in football and the prospects for plenty of splashy plays looks good here.

Purdy is more of a short to mid-range passer with exceptional accuracy and he should be able to orchestrate some scoring drives with the aid of Evans, Kittle and Christian McCaffrey.

The Rams averaged 3.5 offensive touchdowns per game last year while the Niners managed 2.7, while the potential for defensive or special teams scores also make it worthwhile playing the over for this line.

There was very little between the teams when they faced off in Santa Clara last October when the hosts' injury issues were not quite as severe. 

San Francisco pulled out a crucial overtime win on a 41-yard Eddie Pineiro field goal and it would be no surprise if this contest also went down to the wire.

Experts seem split on whether the Niners' early arrival in Australia will give them the edge over the Rams' tried-and-tested 'in-and-out' plan, which helped them to a 35-7 win over Jacksonville Jaguars. 

But there is a greater potential for Los Angeles' gamble to backfire over a journey spanning seven time zones and the main benefit of their schedule may only come when they return stateside after the game.

If LA start sluggishly, it will be tough to recover against a fearsome 49ers defence welcoming back Warner and Bosa.

It is also worth noting that underdogs of 3.5 to 10 points are 61-31 against the spread in Weeks 1 through 3 when the underdog is a team that had a completion percentage of 64% or better the previous year.

The 49ers produced the third-highest completion rate in the NFL of over 68% last year.

Juszcyk featured in the passing game more often against the Rams than other teams last year and could be set for another key role on third downs and in short-yardage situations.

Finally, add over 54.5 points to the Bet Builder. Rams games averaged over 50 points last season and with the 49ers' offence back to full strength, expect them to be pushed all the way in this one.

Niners DT Alfred Collins suffered a torn patellar tendon during the team's first practice in Australia, meaning he joins WR Ricky Pearsall (knee) in missing the entire 2026 season.

TE Kittle should be able to return from the torn Achilles he suffered last season, while DE Bosa looks set to go after recovering from a torn ACL.

WR Evans and RB McCaffrey have missed time while managing injuries leading up to the season but look set to play.

As for the Rams, Garrett is expected to make his team debut in Melbourne despite missing some practice time with a knee injury.

WR Puka Nacua and T Alaric Jackson are expected to play after offseason issues, but head coach Sean McVay announced that Donald will not travel to Australia as he steps up his fitness work after coming out of retirement.
