Accrington are still to win a game this season and sit 22nd in League Two. Meanwhile, after winning promotion last term, Stockport are top of League One.
The pair are in the same pool as Everton U21 and Tranmere.
The match will be broadcast on Sky Sports+.
Accrington Stanley v Stockport County kicks off at 19:45 on Tuesday 3rd September. The game will take place at the Wham Stadium, Accrington.
The pair met twice in League Two last season, with Stockport County winning 3-1 at the Wham Stadium before triumphing 4-2 at Edgeley Park.
Overall, they have won five of the duo's eight meetings, with Accrington Stanley's sole success coming in March 2011.
Accrington sit 22nd in League Two after Saturday's 1-1 home draw against Colchester United, while Stockport are top of League One after Saturday's draw by the same score at Mansfield Town.
