Nicky Henderson's stable star has been out of action since Boxing Day last year after missing the Cheltenham Festival through injury, and was all set to make his long-awaited comeback at Newcastle.
However, Constitution Hill was lame after a gallop at Newbury and could also miss the Christmas Hurdle at Leopardstown.
Henderson has confirmed that Sir Gino will go chasing this season and the unbeaten four-year-old is prominent at the top of the market ahead of the race.
Supreme Novices' Hurdle winner Mystical Power is likely to challenge at the start of what could be a stellar campaign, while Brentford Hope's odds have shortened after finishing second in the Elite Hurdle.
